This is a working overview of peptidase cleavage, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-06-25 and is reviewed periodically as new material appears.
Identity and purity are confirmed with reversed-phase high-performance liquid chromatography, typically monitored at 214 nanometres where the peptide bond absorbs. Mass spectrometry, either electrospray or MALDI-TOF, verifies molecular mass against the theoretical value and detects truncation or adduct formation. Amino acid analysis and peptide mapping provide additional confirmation when required. The most frequently reported impurities are deletion sequences from incomplete coupling, methionine sulfoxide from oxidation, and dimeric species formed through non-covalent aggregation. Impurity profiles depend strongly on the synthesis and purification route chosen by the producer.
Published research has focused mainly on neurological and cognitive endpoints in animal models, with proposed mechanisms involving brain-derived neurotrophic factor and related signalling pathways. A substantial share of the human data originates from a limited number of research groups, and independent replication in other countries remains sparse. Regulatory status reflects that distribution: the peptide is registered as a medicine in Russia and appears in some neighbouring markets, while elsewhere it is handled as a research chemical without approved therapeutic labelling. Questions about dose-response relationships, long-term effects, and comparability across studies are still open.

Circulation time for the peptide is short because peptidases cleave it readily. The Pro-Gly-Pro tail is thought to slow breakdown compared with the bare ACTH fragment, but the gain appears modest. Absorption after intranasal dosing is limited, and only a fraction of a dose is expected to reach the central nervous system. Laboratory concentrations therefore sit well above levels achieved systemically, a gap that complicates translation from bench findings to clinical claims.
The mechanisms attributed to semax are inferred from animal and cell studies rather than traced to one confirmed target. The most frequently cited pathway involves increased expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal and cortical tissue. Some work points to engagement of melanocortin receptors, particularly MC4, which the parent ACTH fragment can activate. Effects on dopaminergic and serotonergic signalling have also been reported. No single account explains all observed results, and the relative weight of each pathway remains unsettled.
Verification of a supplied batch generally combines a certificate of analysis with independent testing, because certificates are self-reported documents. A typical package includes a chromatographic trace, a mass spectrum, and a stated water or counter-ion content. Batch-to-batch consistency matters more than a single purity figure when results are compared across experiments. No single mandatory standard governs research-grade peptide release, so laboratories are expected to define their own acceptance criteria. Residual trifluoroacetate from purification is a frequently overlooked counter-ion.
Lyophilized material is chemically stable for extended periods when kept dry, cold, and protected from light. The powder is hygroscopic, so vials should be warmed to room temperature before opening to reduce condensation on the contents. Once dissolved, the peptide is far less stable because peptide bonds are susceptible to hydrolysis and the methionine residue can oxidize. Solutions are typically aliquoted and held at 2-8 °C for short intervals or frozen for longer ones, and repeated freeze-thaw cycles should be avoided.
Routine characterization relies on reversed-phase high-performance liquid chromatography to establish purity and on mass spectrometry to confirm molecular identity. Electrospray ionization and matrix-assisted laser desorption ionization are both used for mass verification. Amino acid analysis and peptide mapping can detect sequence errors. Common impurities include truncated sequences, methionine sulfoxide formed by oxidation, and deamidated products. Chromatograms are usually recorded near 214 nm, where the peptide backbone absorbs, and purity is reported as the percentage area of the principal peak.
==== European Union ==== In February 2016, orphan designation (EU/3/16/1615) was granted by the European Commission to Gilead Sciences International Ltd, United Kingdom, for remdesivir for the treatment of Ebola virus disease. In April 2020, the European Medicines Agency (EMA) provided recommendations on compassionate use of remdesivir for COVID‑19 in the EU. In May 2020, the Committee for Medicinal Products for Human Use (CHMP) of the EMA recommended expanding the compassionate use of remdesivir to those not on mechanical ventilation. In addition to those undergoing invasive mechanical ventilation, the compassionate use recommendations cover the treatment of hospitalized individuals requiring supplemental oxygen, non-invasive ventilation, high-flow oxygen devices or ECMO (extracorporeal membrane oxygenation). The updated recommendations were based on preliminary results from the NIAID-ACTT study, which suggested a beneficial effect of remdesivir in the treatment of hospitalized individuals with severe COVID‑19. In addition, a treatment duration of five days was introduced alongside the longer ten-day course, based on preliminary results from another study (GS-US-540-5773) suggesting that for those not requiring mechanical ventilation or ECMO, the treatment course may be shortened from ten to five days without any loss of efficacy. Individuals who receive a five-day treatment course but do not show clinical improvement will be eligible to continue receiving remdesivir for an additional five days.
=== Vaccines === Vaccines are an essential part of the response to reduce AMR as they prevent infections, reduce the use and overuse of antimicrobials, and slow the emergence and spread of drug-resistant pathogens. Microorganisms usually do not develop resistance to vaccines because vaccines reduce the spread of the infection and target the pathogen in multiple ways in the same host and possibly in different ways between different hosts. Furthermore, if the use of vaccines increases, there is evidence that antibiotic resistant strains of pathogens will decrease; the need for antibiotics will naturally decrease as vaccines prevent infection before it occurs. A 2024 report by WHO finds that vaccines against 24 pathogens could reduce the number of antibiotics needed by 22% or 2.5 billion DDD every year. If vaccines could be rolled out against all the evaluated pathogens, they could save a third of the hospital costs associated with AMR. Vaccinated people have fewer infections and are protected against potential complications from secondary infections that may need antimicrobial medicines or require admission to hospital. However, there are well documented cases of vaccine resistance, although these are usually much less of a problem than antimicrobial resistance. Antistaphylococcal vaccines have shown limited efficacy, because of immunological variation between Staphylococcus species, and the limited duration of effectiveness of the antibodies produced. Development and testing of more effective vaccines is underway.
=== Trophozoite stage === The trophozoite stage is the infective phase for humans, during which the organism can actively feed and replicate. The trophozoite attaches to the olfactory epithelium, follows the axons of olfactory receptor neurons through the cribriform plate in the nasal cavity, and enters the brain. This reproductive stage of the protozoan organism transforms around 25 °C (77 °F), and thrives best around 42 °C (108 °F), multiplying through binary fission. Trophozoites are characterized by a nucleus surrounded by a flexible membrane. They move via pseudopodia, extending parts of their cell membrane (pseudopods) and filling them with protoplasm to facilitate locomotion. Pseudopods form in the direction of movement. In their free-living state, trophozoites feed on bacteria. In tissues, they appear to phagocytize (enclose and digest) red blood cells and cause tissue damage either through the release of cytolytic substances or by direct cell-to-cell contact using cytolytic membrane proteins. As trophozoites, N. fowleri may develop about one to 12 structures on their membrane known as amoebastomes, also referred to as "suckers" or "food cups", which they use for feeding via a trogocytosis-like mechanism.
== Antibiofilm properties == Cecropin A can destroy planktonic and sessile biofilm-forming uropathogenic E. coli (UPEC) cells, either alone or when combined with the antibiotic nalidixic acid, synergistically clearing infection in vivo (in the insect host Galleria mellonella) without off-target cytotoxicity. The multi-target mechanism of action involves outer membrane permeabilization followed by biofilm disruption triggered by the inhibition of efflux pump activity and interactions with extracellular and intracellular nucleic acids.
=== Postmortem and forensic === After death, the vitreous resists putrefaction longer than other body fluids. Within the hours, days and weeks after death, the vitreous potassium concentration rises, at such a predictable speed that vitreous potassium levels are frequently used to estimate the time since death (post-mortem interval) of a corpse. The metabolic exchange and equilibration between systemic circulation and vitreous humour is so slow that vitreous humour is sometimes the fluid of choice for postmortem analysis of glucose levels or substances which would be more rapidly diffused, degraded, excreted or metabolized from general circulation. According to Judaism, extracting the vitreous fluid for forensic chemical analysis is preferred to blood analysis (in case a forensic or post-mortem toxicology test is deemed necessary). This avoids the loss of even a few droplets of blood from the body prior to burial.

=== Other animals === GBS also causes severe epidemics in farmed fish, causing sepsis and external and internal hemorrhages, having been reported from wild and captive fish involved in epizootics in many countries. Vaccination is an effective method to prevent pathogenic diseases in aquaculture, and different kinds of vaccines to prevent GBS infections have been developed recently. GBS has also been found in many other animals, such as camels, dogs, cats, crocodiles, seals, elephants, and dolphins.
=== Excessive sweating === Botulinum toxin (BTO) injections are a modern clinical trend to treat hyperhidrosis, of which palmar hyperhidrosis is a frequent symptom. For example, AbobotulinumtoxinA has been approved for the treatment of axillary hyperhidrosis, which cannot be managed by topical agents.
=== Impact on negotiations of extraditing suspects to Singapore === In 2002, Singapore tried to negotiate with Australia for the extradition of a British murder suspect and fugitive Michael McCrea, who was wanted for the double murder of a couple whose corpses were discovered abandoned in a car at Orchard Towers. However, McCrea, who was arrested in Australia, was not extradited as Australia, which abolished the death penalty for all offences by then, was not legally allowed to extradite suspects back to countries where they would face the death sentence. It was only after Singapore gave the Australian government the assurance that McCrea would not be hanged even if he was convicted of murder, which allowed McCrea to be sent back to Singapore for trial. McCrea was eventually convicted of culpable homicide and destroying evidence of a murder case, and sentenced to a total of 24 years in jail. This left an impact and precedent on Singapore's avenues to successfully negotiating for extradition of suspects from countries where the death penalty or caning was not practised, including the extradition of suspected bank robber David James Roach, whom the Singapore government promised would not face caning for robbery. Roach was eventually sent back to Singapore, where he later served five years in prison, and he was pardoned from caning by President Halimah Yacob.

The second modification required is the addition of an azide to the N-terminus. Unlike the addition of the alkyne this can be done on the whole peptide, or solely on the N-terminal residue which is added to the rest of the oligopeptide by solid-phase peptide synthesis. The addition of the azide occurs by Cu(II)-catalyzed diazo transfer.

Reversed-phase HPLC gives the main purity figure, most often with UV detection near 214 nanometres. Mass spectrometry then confirms the molecular mass. Together the two methods distinguish a correct sequence from a closely related impurity.
The main chemical risks are methionine oxidation and backbone hydrolysis in solution. Moisture and repeated temperature cycling accelerate both processes. Dry powder held cold and desiccated is considerably more stable than any reconstituted preparation.
It holds a medicine registration in Russia, where it has been used clinically for decades. In most other countries it is treated as a research chemical. That split explains the uneven distribution of clinical literature.
电喷雾或基质辅助激光解吸电离质谱是常用手段,测得的质子化离子质荷比可换算为分子量。结果与理论值落在合理误差内,才支持结构一致的判断。
