A practical reference on Methionine oxidation: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.

Routine characterization relies on reversed-phase high-performance liquid chromatography to establish purity and on mass spectrometry to confirm molecular identity. Electrospray ionization and matrix-assisted laser desorption ionization are both used for mass verification. Amino acid analysis and peptide mapping can detect sequence errors. Common impurities include truncated sequences, methionine sulfoxide formed by oxidation, and deamidated products. Chromatograms are usually recorded near 214 nm, where the peptide backbone absorbs, and purity is reported as the percentage area of the principal peak.
Verification of a supplied batch generally combines a certificate of analysis with independent testing, because certificates are self-reported documents. A typical package includes a chromatographic trace, a mass spectrum, and a stated water or counter-ion content. Batch-to-batch consistency matters more than a single purity figure when results are compared across experiments. No single mandatory standard governs research-grade peptide release, so laboratories are expected to define their own acceptance criteria. Residual trifluoroacetate from purification is a frequently overlooked counter-ion.
Development took place during the 1980s at institutes of the Russian Academy of Sciences, where peptide fragments of ACTH were screened for cognitive and neuroprotective effects. The compound received regulatory approval in Russia as a nasal preparation, marketed for conditions such as ischemic stroke, transient ischemic attacks, and optic nerve disorders. Registration in other countries has not followed. Most published clinical reports originate from a small number of Russian research groups, and independent replication outside that setting remains limited. The regulatory status therefore differs sharply between Russia and the rest of the world.
Laboratory work points toward modulation of neurotrophic signaling, particularly expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal tissue. Studies also describe effects on monoamine turnover, inflammatory mediators, and oxidative markers. These observations come mainly from animal models and cultured cells, so the causal chain in humans is not firmly established. Whether the reported molecular changes translate into measurable clinical benefit is an open question. Reviews generally present the mechanism as plausible rather than demonstrated.
Clinical evidence consists largely of small trials with modest sample sizes, often without independent replication. Reported endpoints include cognitive scores, recovery after stroke, and visual function, but study designs vary widely and few trials meet contemporary reporting standards. Systematic reviewers have noted a high risk of bias in several of these reports. No large multicenter trial conducted outside Russia has been published. The compound is therefore best described as investigational in most jurisdictions, with its clinical role still unresolved.
| Property | Value | Notes |
|---|---|---|
| Typical purity | 95% or higher | By reversed-phase HPLC area |
| Identity method | Electrospray mass spectrometry | Confirms molecular mass |
| Purity method | Reversed-phase HPLC | Detection near 214 nm |
| Primary degradation | Methionine oxidation | Sulfoxide product, +16 Da |
| Solution storage | 2-8 °C, short term | Aliquot to avoid freeze-thaw |
Practical handling follows from those properties. Bulk material is best divided into single-use portions soon after receipt, because each thaw exposes the whole container to moisture and temperature cycling. Vials should be allowed to reach room temperature before opening to prevent condensation on the powder. Low-binding plasticware reduces loss of dilute solutions, and sterile filtration is used when a preparation must remain free of microbial growth. Records of batch number, reconstitution date and storage history are what allow a later analytical result to be interpreted meaningfully.
Identity and purity of Semax are established mainly by reversed-phase high-performance liquid chromatography coupled with mass spectrometry. The chromatographic trace gives a purity estimate as a percentage of total peak area, while electrospray or matrix-assisted laser desorption ionization confirms the molecular mass against the calculated value. Amino acid analysis and sequence-specific fragmentation provide further confirmation when a supplier's chain of custody is unclear. Vendors frequently quote a purity figure without stating the detection wavelength or the integration method, which limits how far one number can be compared with another.
The parent fragment ACTH(4-10) carries the sequence Met-Glu-His-Phe-Arg-Trp-Gly. Semax replaces the arginine and tryptophan positions with a proline-glycine-proline tail, giving Met-Glu-His-Phe-Pro-Gly-Pro. That change removes residues associated with adrenal stimulation, so the peptide does not drive cortisol release the way full ACTH does. This distinction shapes how the compound is grouped in the literature, where it sits with neuropeptides and peptide neuromodulators rather than with corticosteroids.
Regulatory status varies sharply by country. Semax is registered for medical use in Russia, where it appears in formularies as a nasal solution, and it also holds registration in a small number of neighbouring states. It has no approval from the United States Food and Drug Administration or the European Medicines Agency, and it is not a scheduled controlled substance in most jurisdictions. Elsewhere it circulates mainly as laboratory material, so purity documentation comes from suppliers rather than from a national pharmacopoeia.
Semax is a synthetic peptide created in the Soviet Union during the early 1980s by researchers working in Moscow. It was built from the short adrenocorticotropic hormone fragment known as ACTH(4-10), and the chain was then extended with three additional amino acids. The resulting molecule was named semax and entered clinical use in Russia in 1994. It is generally described as a nootropic and neuroprotective agent rather than as a hormone analogue.
Formaldehyde ( for-MAL-di-hide, US also fər-) (systematic name methanal) is a pungent colorless gas mainly used in the production of industrial resins, such as for particle board, coatings, plastics, pulp, paper, synthetic fibers, and textiles. A ~5% solution in water of formaldehyde is used as a disinfectant and fumigant in industrial, agricultural, and healthcare settings, and a ~37% solution is used to preserve tissue samples in labs. In 2024, the global production of formaldehyde was estimated at 26 million tons per year, and is a precursor to many other materials and chemical compounds. It is an organic compound with the chemical formula CH2O and structure H2C=O. The gas spontaneously polymerizes into paraformaldehyde. It is stored as a ~37% aqueous solution known as formalin, which consists mainly of the hydrate CH2(OH)2. It is the simplest of the aldehydes (R−CHO). Formaldehyde also occurs naturally. It is derived from the degradation of serine, dimethylglycine, and lipids. Demethylases act by converting N-methyl groups to formaldehyde. Formaldehyde is classified as a group 1 carcinogen and can cause respiratory and skin irritation upon exposure.
=== By individual compound === This list includes notable compounds representative of most of the structural variations that have been explored in this series, but is by no means exhaustive. Many derivatives invented for scientific study into the structure-activity relationships of 5-HT2 receptor agonists have never appeared as designer drugs, while conversely some derivatives that have appeared as designer drugs are structurally novel and of unknown pharmacological activity (e.g. C30-NBOMe, 5-APB-NBOMe).
Juan Vucetich, an Argentine chief police officer, created the first method of recording the fingerprints of individuals on file. In 1892, after studying Galton's pattern types, Vucetich set up the world's first fingerprint bureau. In that same year, Francisca Rojas of Necochea was found in a house with neck injuries whilst her two sons were found dead with their throats cut. Rojas accused a neighbour, but despite brutal interrogation, this neighbour would not confess to the crimes. Inspector Alvarez, a colleague of Vucetich, went to the scene and found a bloody thumb mark on a door. When it was compared with Rojas' prints, it was found to be identical with her right thumb. She then confessed to the murder of her sons. A Fingerprint Bureau was established in Calcutta (Kolkata), India, in 1897, after the Council of the Governor General approved a committee report that fingerprints should be used for the classification of criminal records. Working in the Calcutta Anthropometric Bureau, before it became the Fingerprint Bureau, were Azizul Haque and Hem Chandra Bose. Haque and Bose were Indian fingerprint experts who have been credited with the primary development of a fingerprint classification system eventually named after their supervisor, Sir Edward Richard Henry. The Henry Classification System, co-devised by Haque and Bose, was accepted in England and Wales when the first United Kingdom Fingerprint Bureau was founded in Scotland Yard, the Metropolitan Police headquarters, London, in 1901. Sir Edward Richard Henry subsequently achieved improvements in dactyloscopy.
== History == Efficacy was evaluated in AGAVE-201 (NCT04710576), a randomized, open-label, multicenter trial investigating three dosages of axatilimab in people with recurrent or refractory chronic graft-versus-host disease who had received at least two lines of systemic therapy and required additional treatment. Axatilimab was evaluated in an open-label single arm clinical trial of 79 participants with chronic graft-versus-host disease who had received at least two prior systemic treatments and required additional treatment. All participants received axatilimab 0.3 mg/kg every two weeks, until chronic graft-versus-host disease progression or unacceptable toxicity. The US Food and Drug Administration (FDA) approved axatilimab based on evidence of safety and efficacy from a clinical trial which included a total of 79 participants with chronic graft-versus-host disease after failure of two prior lines of systemic therapy. The trial was conducted at 55 sites in 13 countries, including Australia, Belgium, Canada, Germany, Greece, France, Israel, Italy, Spain, South Korea, Taiwan, the United Kingdom, and the United States. There were 24 sites in the United States. Out of 79 participants, at the approved dose of 0.3 mg/kg every two weeks, 37 participants were enrolled in the United States, and 43 participants at the trial sites outside of the United States. The FDA granted the application for axatilimab orphan drug and fast track designations for the treatment of chronic graft-versus-host disease.
Akt resides in the cytosol in an inactive conformation, until the cell is stimulated and it translocates to the plasma membrane. The Akt PH domain has a high affinity for second messenger PI(3,4,5)P3, binding to it preferentially over other phosphoinositides. Thus PI3K activity is essential for translocation of Akt to the membrane. Interaction with PI(3,4,5)P3 causes conformational changes and exposure of phosphorylation sites Thr308 in the kinase domain and Ser473 in the C-terminal domain. Akt is partially activated by phosphorylation of T308 by PDK1. Full activation requires phosphorylation of S473, which can be catalysed by multiple proteins, including phosphoinositide-dependent kinase 2 (PDK2), integrin-linked kinase (ILK), mechanistic target of rapamycin complex complex 2 (mTORC2) and DNA-dependent protein kinase (DNA-PK). The regulation of Ser473 phosphorylation is not fully understood but may also be influenced by autophosphorylation after Thr308 phosphorylation. After stimulation, the levels of PIP3 decrease and Akt activity is attenuated by dephosphorylation by serine/threonine phosphatases.

Linked-read sequencing can facilitate de novo genome assembly, which involves reconstructing a genome from scratch without any prior reference. Linked-read sequencing enables assembly of large genomic regions, and helps improve the completeness and contiguity of the resulting genome. This can be particularly useful for studying organisms that lack a high-quality reference genome, such as non-model organisms or organisms with complex genomes. Many scientists have been using linked-read sequencing technology for de novo genome assembly recently in a variety of organisms, including humans, plants, and animals. For example, Dr. Evan Eichler and his research group used linked-read sequencing to assemble genome of orangutan, which had previously been difficult to study due to its complex genome. The resulting genome assembly helped scientists to study new insights into the evolutionary history of primates and the genetic basis of human diseases. Also, the aligned or assembled reads can be used for other genetic investigations or downstream analysis, such as haplotype phasing.

== Early life and education == Hashim Thaçi was born on 24 April 1969 in the village of Burojë, Skenderaj, SFR Yugoslavia (now Kosovo). His family comes from the Thaçi tribe. Skenderaj is located in the Drenica valley, a region historically resistant to Serbian rule. Drenica would become the birthplace of the Kosovo Liberation Army (KLA) in the early 1990s. Thaçi studied philosophy and history at the University of Pristina. By 1993, he was living in Switzerland, where he joined the Albanian political émigré group. He registered for postgraduate studies at the University of Zürich in the departments of history and international relations, achieving a master's degree. As a young man, Thaçi joined the People's Movement of Kosovo (LPK), an underground group committed to overthrowing the Yugoslav government that had been historically financed and ideologically shaped by the regime of Albania's former dictator, Enver Hoxha.

A¹ – 2-aminoisobutyric acid (Aib). L² – leucine modified with an α-methyl substituent (MeL, 2-methylleucine). S³ – L-serinamide (L-serine with the carboxylic acid group replaced with a carboxamide). K⁴ – L-lysine with the amino group at position 6 modified with a side chain; specifically, (AEEA)-(γ-Glu)-(C20 diacid) (where AEEA is 2-[2-(2-aminoethoxy)ethoxy]acetic acid, commonly used as a spacer group in synthetic peptides). Retatrutide is synthetically engineered and produced using solid-phase peptide synthesis (SPPS). This is when amino acids are added to a peptide chain and attached to solid resin forming a backbone. After the backbone is synthesized, the molecule is cleaved chemically, then purified. Lipidation modifications also occur, meaning a fatty-acid side chain is added to promote reversible binding to albumin. This modification allows for a longer drug half-life, raising compliance as it enables once-weekly dosing.
In a survey of the general population, it was found that although many erect penises point upwards, it is common and normal for the erect penis to point nearly vertically upwards or horizontally straight forward or even nearly vertically downwards, all depending on the tension of the suspensory ligament that holds it in position. 37% of healthy men have a penile curve. With 22.2% of men having upwards curvature and 14.8% of men having downwards curvature.
Approval by the US Food and Drug Administration followed in December 1993, also for use as an adjuvant (effective when added to other antiseizure drugs) medication to control partial seizures in adults; that indication was extended to children in 2000. Subsequently, gabapentin was approved in the United States for the treatment of pain from postherpetic neuralgia in 2002. A generic version of gabapentin first became available in the United States in 2004. An extended-release formulation of gabapentin for once-daily administration, under the brand name Gralise, was approved in the United States for the treatment of postherpetic neuralgia in January 2011. Gabapentin has been prescribed for a variety of disorders and is one of the more common medications used, particularly in elderly people.
Terlipressin, sold under the brand name Terlivaz among others, is an analogue of vasopressin used as a vasoactive drug in the management of low blood pressure. It has been found to be effective when norepinephrine does not help. Terlipressin is a vasopressin receptor agonist. Terlipressin was approved for medical use in the United States in 2022. The US Food and Drug Administration (FDA) considers it to be a first-in-class medication.

Dry powder is normally held at -20 °C or lower, away from light and moisture. Sealed vials can also be kept at 2-8 °C for shorter intervals. Warming to room temperature before opening prevents condensation.
In solution, hydrolysis of peptide bonds and oxidation of the methionine residue are the principal concerns. Oxidation converts methionine to its sulfoxide form, shifting the mass by sixteen daltons. Both processes are slowed by cold storage and by avoiding alkaline conditions.
A certificate documents what the supplier measured on a specific lot, not how a laboratory will use the material. Independent verification of identity and purity is common practice. Acceptance criteria should be defined before a batch is ordered.
Semax is a synthetic seven-amino-acid peptide derived from a fragment of corticotropin. It is used in Russia as a nasal preparation, while elsewhere it is studied as a research compound. It is not approved as a medicine in most countries.
