Endochondral ossification, or intracartilaginous ossification, is the formation of long bones and other bones. This requires a hyaline cartilage precursor. There are two centers of ossification for endochondral ossification. The primary center In long bones, bone tissue first appears in the diaphysis (middle of shaft). Chondrocytes multiply and form trabeculae. Cartilage is progressively eroded and replaced by hardened bone, extending towards the epiphysis. A perichondrium layer surrounding the cartilage forms the periosteum, which generates osteogenic cells that then go on to make a collar that encircles the outside of the bone and remodels the medullary cavity on the inside. The nutrient artery enters via the nutrient foramen from a small opening in the diaphysis. It invades the primary center of ossification, bringing osteogenic cells (osteoblasts on the outside, osteoclasts on the inside.) The canal of the nutrient foramen is directed away from more active end of bone when one end grows more than the other. When bone grows at same rate at both ends, the nutrient artery is perpendicular to the bone. Most other bones (e.g. vertebrae) also have primary ossification centers, and bone is laid down in a similar manner. Secondary centers The secondary centers generally appear at the epiphysis. === Mechanism of action === Zalcitabine is an analog of pyrimidine. It is a derivative of the naturally existing deoxycytidine, made by replacing the hydroxyl group in position 3' with a hydrogen. It is phosphorylated in T cells and other HIV target cells into its active triphosphate form, ddCTP. This active metabolite works as a substrate for HIV reverse transcriptase, and also by incorporation into the viral DNA, hence terminating the chain elongation due to the missing hydroxyl group. Apart from inhibiting retroviral reverse transcriptase, ddC inhibits mitochondrial DNA polymerase gamma, resulting in a dose-limiting toxicity.

== Causes == The best-known of cerebral folate deficiency is due to a genetic mutation in the FOLR1 gene. It is inherited in an autosomal recessive manner. The mutation of the FOLR1 gene causes an inability to produce the folate receptor alpha (FRA) protein. CFD can also involve the malfunction and disruption of the FRA in other ways. One way the FRA can be disrupted is by the attachment of the autoantibodies, causing dysfunction in the receptor. Also, a mitochondrial disease can impact the functioning of the folate receptor alpha. In order for the receptor to function properly, energy from the mitochondria is required. Folate must be actively transported into the brain, so ATP from the mitochondria is essential. If the individual has a mitochondrial disease, the FRA could be lacking adequate energy, resulting in the deficiency of folate in the brain. Besides a malfunctioning transport system, cerebral folate deficiency can also be caused by malfunctioning biochemical pathways, often mutations in genes that code for enzymes in the folate. This is seen in 5,10-methenyltetrahydrofolate synthetase deficiency (MTHFS deficiency), MTHFR deficiency, DHFR deficiency, and occasionally MTHFD1 defienciency. Sometimes less-related biochemical pathways can also lead to a presentation of CFD, often called a "secondary" cerebral folate deficiency. CFD can develop in AADC deficiency through the depletion of methyl donors, such as SAM and 5-MTHF, by O-methylation of the excessive amounts of L-DOPA present in patients.

== Professional activities == After earning a Ph.D. degree in biochemistry, a medical degree, and completing a research postdoctoral fellowship at the University of Wisconsin–Madison, Holick completed a residency in medicine at the Massachusetts General Hospital in Boston. He has been practicing adult and pediatric endocrinology since 1978 and is professor of medicine, physiology and biophysics and director of the Bone Health Care Clinic and the Heliotherapy, Light, and Skin Research Center at Boston University Medical Center. It provides extensive evaluation and treatment programs for children and adults with various metabolic bone diseases including osteoporosis, osteomalacia, stress fractures in young athletic women and men, and minimum trauma and nontraumatic fractures in infants, children and adults with hypermobility syndromes, osteogenesis imperfecta, and Ehlers–Danlos syndrome. He has been director of the General Clinical Research Unit at Boston University for several years. Deuterated mitragynine (developmental code name KUR-101) is an atypical μ-opioid receptor agonist and deuterated analogue of mitragynine (found in kratom) which is under development for the treatment of acute pain and opioid-related disorders. It is taken by mouth. Deumitragynine is said to have analgesic effects. The drug is being developed by ATAI Life Sciences and Kures. As of December 2022, it is in phase 1 clinical trials for acute pain and opioid-related disorders.

A high main-peak percentage says little about identity, counter-ion content, or residual solvents. It also does not distinguish an isomer or a closely eluting analog. Mass confirmation and the accompanying documentation cover those gaps.
Salt form changes mass, solubility, and hygroscopicity, and acetate and trifluoroacetate salts behave differently in solution. Storage guidance still centers on -20 °C for powder and lower temperatures for solutions. The certificate should state which salt is present.
