===== Vildagliptin ===== Vildagliptin (Galvus)(Figure 6) was first synthesized in May 1998 and was named after Edwin B. Villhauer. It was discovered when researchers at Novartis examined adamantyl derivatives that had proven to be very potent. The adamantyl group worked as a steric bulk and slowed intramolecular cyclization while increasing chemical stability. Furthermore, the primary metabolites were highly active. To avoid additional chiral center a hydroxylation at the adamantyl ring was carried out (Figure 6). The product, vildagliptin, was even more stable, undergoing intramolecular cyclization 30-times slower, and having high DPP-4 inhibitory activity and longer-lasting pharmacodynamic effect.

Journalists interpreted the story as a synopsis of what could have been the plot for Episode Three, or for Borealis, another canceled project led by Laidlaw. Substituting the characters and locations with their Half-Life counterparts, "Epistle 3" has Gordon and Alyx travel to the Arctic to board the Borealis, a ship that travels erratically through time and space, where they confront alternative versions of themselves. They rig the Borealis to travel to the heart of the alien Combine empire and self-destruct. Before it explodes, the mysterious G-Man extracts Alyx and Gordon is rescued by the friendly Vortigaunt aliens. Walker denied that the story had been Valve's plan for Episode Three, and said that it was likely just one of many ideas by Laidlaw. In a 2023 interview, Laidlaw said it was not representative of Episode Three, as "all the real story development can only happen in the crucible of developing the game". He said he regretted publishing it, as it had created problems for his former colleagues at Valve, and that he had been "deranged" and "completely out of touch".

The buffer liquid between the two tubes is at a gradually rising concentration, always a bit over the incoming fluid, in this example reaching 1200 mg/L. This is regulated by the pumping action on the returning tube as will be explained immediately. The tip of the loop has the highest concentration of salt (NaCl) in the incoming tube—in the example 1199 mg/L, and in the buffer 1200 mg/L. The returning tube has active transport pumps, pumping salt out to the buffer liquid at a low difference of concentrations of up to 200 mg/L more than in the tube. Thus when opposite the 1000 mg/L in the buffer liquid, the concentration in the tube is 800 and only 200 mg/L are needed to be pumped out. But the same is true anywhere along the line, so that at exit of the loop also only 200 mg/L need to be pumped. In effect, this can be seen as a gradually multiplying effect—hence the name of the phenomena: a 'countercurrent multiplier' or the mechanism: Countercurrent multiplication, but in current engineering terms, countercurrent multiplication is any process where only slight pumping is needed, due to the constant small difference of concentration or heat along the process, gradually raising to its maximum. There is no need for a buffer liquid, if the desired effect is receiving a high concentration at the output pipe.
== Safety == Leucine toxicity, as seen in decompensated maple syrup urine disease, causes delirium and neurologic compromise, and can be life-threatening. A high intake of leucine may cause or exacerbate symptoms of pellagra in people with low niacin status because it interferes with the conversion of L-tryptophan to niacin. Leucine at a dose exceeding 500 mg/kg/d was observed with hyperammonemia. As such, unofficially, a tolerable upper intake level (UL) for leucine in healthy adult men can be suggested at 500 mg/kg/d or 35 g/d under acute dietary conditions.

DynaLIFE was a privately owned Canadian medical laboratory company based in Edmonton, Alberta. In May 2022, Alberta Health Services (AHS) signed a 15-year contract valued at approximately $4.8 billion with DynaLIFE to provide community laboratory services across Alberta.
